VARIETAS KEDELAI (Glycine max L. Merr) PADA INDUKSI MUTASI
MENGGUNAKAN EMS (Ethyl-methanesulfonate)

ABSTRAK
Kedelai (Glycine max L. Merr) merupakan tanaman sumber protein alternatif.
Keanekaragaman kedelai dapat ditingkatkan dengan induksi mutasi. Penelitian ini
bertujuan untuk mendapatkan interaksi terbaik antara varietas kedelai dan
konsentrasi EMS (Ethyl-methanesulfonate) terhadap morfologi, pertumbuhan dan
hasil tanaman kedelai. Metode penelitian menggunakan Rancangan Acak Lengkap
(RAL) Faktorial, 2 faktor dengan 3 kali ulangan. Faktor pertama konsentrasi EMS
dengan taraf 0 ppm, 250 ppm, 500 ppm dan 750 ppm. Faktor kedua varietas benih
yaitu: varietas Argomulyo, Gepak Kuning dan Detam-1. Data dianalisis dengan
analisis ragam (ANOVA) lalu, diuji lanjut dengan uji DMRT dan uji polinomial
ortogonal taraf 5%. LC50 EMS dihitung menggunakan Ms. Excell dan aplikasi
CurveExpert. Hasil penelitian menunjukkan terdapat respon morfologi pada tipe
pertumbuhan batang. Terdapat interaksi antara konsentrasi EMS dan varietas pada
variabel umur berbunga, jumlah cabang, jumlah polong per tanaman, hasil panen
per petak dan potensi hasil panen per hektar. Konsentrasi EMS 335 ppm optimal
untuk meningkatkan bobot 100 biji. LC50 EMS dicapai pada 878,90 ppm. Varietas
Gepak Kuning memberikan hasil yang lebih baik pada uji perkecambahan,
persentase hidup, jumlah cabang dan jumlah polong per tanaman. Varietas
Argomulyo dan Detam-1 memberikan hasil yang lebih baik pada variabel tinggi
hipokotil dan bobot 100 biji, dan hasil panen per petak.

SOYBEAN VARIETIES (Glycine max L. Merr) ON MUTATION
INDUCTION USING EMS (Ethyl-methanesulfonate)

ABSTRACT
Soybean (Glycine max L. Merr) is an alternative protein. Soybean diversity can be
increased by mutation induction. This research aims to obtain the best interaction
between soybean varieties and EMS (Ethyl-methanesulfonate) concentration on the
morphology, growth and yield of soybean plants. This research used a factorial
Completely Randomized Design (CRD) method, with 2 factors and 3 replications.
The first factor is EMS concentration with 0 ppm, 250 ppm, 500 ppm and 750 ppm
levels. The second factor is soybean varieties: Argomulyo, Gepak Kuning and
Detam-1 varieties. The data were analyzed using Analysis of Variance (ANOVA)
and then continued with DMRT and orthogonal polynomial test at 5% level. LC50
EMS is calculated using Ms. Excell and CurveExpert applications. The results show
that there is a morphological response on growth type and an interaction between
EMS concentration and variety on the variables of flowering age, number of
branches, number of pods per plant, yield per plot and potential yield per hectare.
EMS concentration of 335 ppm is optimal for increasing the weight of 100 seeds.
LC50 EMS was achieved at 878.90 ppm. Gepak Kuning variety has better results
in germination tests, survival percentage, number of branches and number of pods
per plant. Argomulyo and Detam-1 varieties have better results in the variables of
hypocotyl height and 100 seed weight, and yield per plot.
