Abstract

Abstract - Hydrogen-producing bacteria can be found in the activated sludge generated from anaerobik waste treatment processes. Microbes in it consists of acetogenesis, metanogenesis and hydrogen -producing bacteria such as Bacillus and Clostridium. The purpose of this study to isolate the hydrogen -producing microbes from the mix culture in the anaerobik activated sludge. Sludge from the anaerobik bioreaktor simmer for 2 hours to kill vegetative cells so that only spores are present. Then the spores cultured in the hydrogen media in anaerobik conditions, at a temperature of 65o C for 3 times 72 hours. Hydrogen gas generated during the breeding is analized using GC. Identification of the hydrogen producing bacteria is conducted using DNA tests. It’s performed by 16S rRNA gene sequence comparison with the data base available in the GenBank/EMBL/DDBJ / PDB through the Blast program, showed that all three isolates were identified as Bacillus citrulans.
