BERBAGAI KONSENTRASI AUKSIN DAN MEDIA TANAM

ABSTRAK
Penyediaan bibit kentang bermutu merupakan masalah yang dihadapi petani.
Perbanyakan secara kultur jaringan merupakan upaya untuk meningkatkan
ketersediaan bibit bermutu. Keberhasilan kultur jaringan dipengaruhi oleh
kemampuan mengendalikan kondisi lingkungan, zat pengatur tumbuh serta media
tanam pada tahap aklimatisasi. Tujuan penelitian adalah menentukan interaksi
antara penggunaan konsentrasi auksin dengan media tanam serta mendapatkan
konsentrasi auksin dan media tanam yang tepat untuk aklimatisasi kentang.
Penelitian menggunakan Rancangan Petak Terbagi 2 faktor dengan 3 ulangan.
Petak utama adalah konsentrasi auksin terdiri dari 3 taraf; 1 ppm, 2 ppm, dan 3 ppm.
Anak petak adalah media tanam terdiri dari 3 taraf; arang sekam:kascing (1:1),
cocopeat:kascing (1:1), dan arang sekam:cocopeat:kascing (1:1:1). Data dianalisis
menggunakan ANOVA pada taraf 5% dan dilanjutkan dengan DMRT pada taraf
5%. Hasil penelitian menunjukkan terdapat interaksi antara perlakuan konsentrasi
IAA 3 ppm dengan media tanam arang sekam:cocopeat:kascing (1:1:1) pada
parameter tinggi tanaman 4 MST. Konsentrasi IAA 3 ppm lebih baik dibandingkan
konsentrasi 1 ppm pada parameter jumlah daun 8 MST. Media tanam arang
sekam:cocopeat:kascing (1:1:1) lebih baik dibandingkan media tanam arang
sekam:kascing (1:1) pada parameter tinggi tanaman 8 MST, jumlah daun 8 MST,
panjang daun dan lebar daun 8 MST, serta bobot segar tanaman.

POTATO (Solanum tuberosum L.) PLANLET ACCLIMATIZATION ON
VARIOUS AUXIN CONCENTRATIONS AND GROWING MEDIA

ABSTRACT
Provision of quality potato seeds is a problem faced by farmers. Tissue culture
propagation is an effort to increase the availability of quality seeds. The success of
tissue culture is influenced by the ability to control environmental conditions,
growth regulators and growing media at the acclimatization stage. The aim of the
research was to determine the interaction between the use of auxin concentrations
and the planting medium and to obtain the appropriate concentration of auxin and
planting medium for potato acclimatization. The research method used a split plot
design with 2 factors and 3 replications. The main plot is the auxin concentration
consisting of 3 levels; 1 ppm, 2 ppm and 3 ppm. Subplots are planting media
consisting of 3 levels; husk charcoal:vermicompost (1:1), cocopeat:vermicompost
(1:1), and husk charcoal:cocopeat:vermicompost (1:1:1). Data were analyzed using
ANOVA at 5% level and followed by DMRT at 5% level. The results showed that
there was an interaction between the treatment of 3 ppm IAA concentration and the
planting medium of husk charcoal:cocopeat:vermicompost (1:1:1) on the plant
height parameter 4 WAP. The concentration of 3 ppm IAA was better than the
concentration of 1 ppm on the parameter number of leaves 8 WAP. The planting
medium of husk charcoal:cocopeat:vermicompost (1:1:1) media was better than
husk charcoal:vermicompost on the parameters of plant height 8 WAP, number of
leaves 8 WAP, leaf length and width 8 WAP, and plant fresh weight.
