Abstract

Nephentes termasuk salah satu tanaman hias tropis di Indonesia yang mulai
terancam punah. Salah satu cara melestarikan nephentes agar tidak terancam
punah yaitu dengan menggunakan perbanyakan dengan Teknik kultur jaringan.
Penelitian ini bertujuan untuk menentukan interaksi konsentrasi IAA dan
thiamin yang paling baik terhadap mikrostek Nephentes secara in vitro,
menentukan konsentrasi IAA yang paling baik terhadap mikrostek Nephentes
secara in vitro, dan menentukan konsentrasi thiamin yang paling baik terhadap
mirkostek Nephentes secara in vitro. Penelitian ini dilaksanakan
di Laboratorium Bioteknologi Fakultas Pertanian Universitas Pembangunan
Nasional “Veteran” Yogyakarta pada bulan November 2020 sampai Februari
2021. Metode penelitian yang digunakan adalah percobaan laboratorium
dengan rancangan lingkungan Rancangan Acak Lengkap (RAL) dengan dua
faktor. Faktor pertama adalah konsentrasi IAA 1,5 mg/l, 2 mg/l dan 2,5 mg/l.
Faktor kedua adalah konsentrasi thiamin 10 mg/l, 11 mg/l dan 12 mg/l. Setiap
kombinasi diulang 3 kali. Hasil penelitian menunjukkan bahwa terdapat
interaksi antara konsentrasi IAA dan thiamin, kombinasi paling baik adalah
pada kombinasi perlakuan I2T2 (IAA 2mg/l dan thiamin 11 mg/l) pada
parameter tinggi planlet, dan kombinasi perlakuan I3T3 (IAA 2,5 mg/l dan
thiamin 12 mg/l) pada parameter jumlah daun. Konsentrasi IAA 1,5 mg/l
merupakan konsentrasi paling baik pada parameter jumlah tunas, bobot segar
dan jumlah kantong bagi pertumbuhan mikrostek kantong semar secara in vitro.
Konsentrasi thiamin 11 mg/l merupakan konsentrasi paling baik pada parameter
bobot kering planlet bagi pertumbuhan mikrostek kantong semar secara in vitro.

WITH ADDITION OF IAA AND THIAMIN BY IN VITRO

ABSTRACT
Nephentes is one of the tropical ornamental plants in Indonesia which is starting to be
threatened with extinction. One way to preserve nephentes so that they are not
threatened with extinction is by using tissue culture techniques. This study aimed to
determine the best interaction of IAA and thiamin concentrations against micro cuttings
propagation of Nephentes by in vitro, determine the best IAA concentration against
micro cuttings propagation of Nephentes by in vitro, and determine the best
concentration of thiamin against micro cuttings propagation of Nephentes by in vitro.
This research was conducted in Biotechnology Laboratorium Of UPN Veteran
Yogyakarta on November 2020 to February 2021. This research was laboratory
experiment with completely randomized design in two factors. The first factor is
concentration of thiamin 10 mg/l, 11 mg/l and 12 mg/l and the second factor is
concentration of IAA 1,5 mg/l, 2 mg/l and 2,5 mg/l. Each treatment combination was
repeated 3 times. The obtained data was analyzed using analysis of variance (ANOVA)
, and if a significant diferrence effect existed, it would be followed by DMRT
significant difference at level 5%. The result shown that there is an interaction of the
treatment combination I2T2 (IAA 1 mg/l and thiamin 11 mg/l) on the parameter of
height of the planlet, and treatment combination A3B3 (IAA 2,5 mg/l and thiamin 12
mg/l) on the parameter of number of leaves. The use of IAA 1,5 mg/l gave the best
result in live percentage, number of shoots, planlet fresh weight and numbers of bags.
The use of thiamin 11 mg/l gave the best result in planlet dry weight.
