Abstract
Abstract Mangostees (Garcinia mangostana L.) is a very beneficial tropical fruit. Its proliferation can be done with in vitro method. The purpose of this research is for determining the exact combination between the number of parts of the grain and the kind of natural media for proliferation mangosteen bud using in vitro culture. The research was held in Laboratorium Kultur Jaringan Fakultas Pertanian UPN “Veteran” Yogyakarta, on April until November 2015. The materials are Puspahiang mangosteen seeds, green bean sprouts extract, coconut water, chemical, and tools for plant tissue isolation. The experiment was designed factorially (3x4). Factor I was grains treatment (3 degrees) which are B1: whole, B2: two-split, B3: four split. Factor II was natural media treatment (4 degrees) which are M1:MS + coconut water 25%, M2:MS + coconut water 75%, M3:MS + green bean sprouts extract 25%, and M4: MS + green bean sprouts extract 75%. Every experiment unit was arranged in Complete Random Design with ten replications. For temporary information, the contamination time was faster (4 days after incubation) compared to green bean sprouts extract on MS + 75% coconut water media, the highest contamination was 76,7%. Keywords: mangosteen, natural media, in vitro Abstrak Manggis (Garcinia mangostana L.) adalah jenis buah tropik yang manfaatnya sangat luas. Perbanyakannya dapat dilakukan secara in vitro. Tujuan penelitian untuk menetukan kombinasi yang tepat antara jumlah belahan biji dan macam media alami untuk perbanyakan/proliferasi tunas manggis secara kultur in vitro. Penelitian dilaksanakan di Laboratorium Kultur Jaringan Fakultas Pertanian UPN “Veteran” Yogyakarta, pada bulan April hingga Nopember 2015. Bahan berupa biji buah manggis varietas Puspahiang, media MS, ekstrak touge kacang hijau, air kelapa, bahan kimia dan peralatan pendukung kultur jaringan. Percobaan dirancang secara faktorial (3x4). Faktor I. adalah perlakuan biji belah (3 taraf) yaitu B1: utuh, B2: belah dua, dan B3: belah empat. Faktor II. adalah perlakuan media alami (4 taraf) yaitu M1: MS + air kelapa 25 %, M2: MS + air kelapa 75 %, M3: MS + ekstrak touge kacang hijau 25 %, dan M4: MS + ekstrak touge kacang hijau 75 %. Tiap unit percobaan disusun dalam Rancangan Acak Lengkap dengan 10 ulangan. Diinformasikan sementara bahwa pada media MS + air kelapa 75%, waktu munculnya bakteri (terkontaminasi) nyata lebih cepat (4 hari setelah inkubasi) dibandingkan dengan media ekstrak tauge kacang hijau dan jumlah yang terkontaminasi nyata tertinggi yaitu 76,7 %.
